Characteristics of Lactic Acid Bacteria Isolated from Traditional Wheat Nuruk and their Effects on Nitric Oxide Production in Mouse Macrophage

Abstract

|
The purpose of this study is to assess of efficacy about lactic acid bacteria isolated form traditional wheat Nuruk. 6 strains was isolated from traditional Nuruk. The Strains identified by 16s rRNA gene sequencing are Lactobacillus Curvatus (named SA-13, YS-2), Pediococcus Acidilactici (named SS-5, YS-3) and Pediococcus Pentosaceus (named SA-2, YS-1). P. acidilactici SS-5 has Acid and bile tolerance. The bacteria free - supernatant of P. acidilactici SS-5 showed antibacterial activity in gram negative bacteria (Escherichia Coli, Salmonella Typhimurium) and gram positive bacteria (Staphylococcus Aureus, Listeria Monocytogenes) in paper disk diffusion assay. The bacteria free - supernatant of P. acidilactici SS-5 showed effect in all of used pathogenic bacteria at concentration of 11.25 mg/mL. All strains isolated from traditional Nuruk showed more than 60 percents on the antioxidant activity. Raw 264.7 cell line was used as a macrophage model to assess the effects on cytotoxic test and the production of nitric oxide. The nitric oxide productions of the viable bacteria and heat-inactivated bacteria of the P. acidilactici SS-5 were superior to other strains isolated Nuruk.
